What is printed. The shirt carries the skeletal formula of serotonin, known to chemists as 5-hydroxytryptamine or 5-HT, drawn as a line diagram with the name and the formula C10H12N2O. The structure is an indole, a benzene ring fused to a five-membered ring containing nitrogen, with a hydroxyl group on the benzene side and a short aminoethyl chain on the other. The same indole core appears in the amino acid tryptophan, from which serotonin is made, and in psilocybin, DMT and LSD. Serotonin is a neurotransmitter and a local hormone. About ninety percent of it in the human body is not in the brain at all but in the lining of the gut.

History

Serotonin was discovered twice, in two organs, under two names. In 1937 the Italian pharmacologist Vittorio Erspamer, working in Pavia, extracted a substance from the enterochromaffin cells of the gut lining that made smooth muscle contract; he showed it was an amine and called it enteramine. Independently, at the Cleveland Clinic in 1948, Maurice Rapport, Arda Green and Irvine Page isolated a vasoconstricting factor from blood serum that had been getting in the way of their hypertension research. They named it serotonin, from serum and tonic. In 1949 Rapport worked out its structure as 5-hydroxytryptamine, and in 1951 chemists at Abbott and at Upjohn reported syntheses of the pure compound, which made it available for experiments everywhere. In 1952 Erspamer and Asero confirmed that enteramine and serotonin were the same molecule. The next year Betty Twarog, a young biochemist in Page's laboratory, detected serotonin in the brains of dogs, rats and rabbits, the finding that moved it from blood chemistry into neuroscience. Also in 1953, John Gaddum in Edinburgh showed that LSD blocks serotonin's action on smooth muscle, which seeded the idea that the molecule had something to do with mood and perception. In 1957 Gaddum and Picarelli distinguished the first two receptor types pharmacologically and named them D and M; today fourteen subtypes are known.

What serotonin does

Serotonin acts on at least fourteen receptor subtypes grouped into seven families, 5-HT1 through 5-HT7. All are G protein-coupled receptors except 5-HT3, which is a ligand-gated ion channel. Most of the body's serotonin is made by enterochromaffin cells of the intestine, where it regulates gut motility and secretion. Platelets take it up from the blood and release it at sites of injury to constrict vessels and promote clotting. In the brain, serotonin neurons are concentrated in the raphe nuclei of the brainstem and project widely, modulating sleep, appetite, mood and sensory processing. Serotonin is synthesized from tryptophan in two enzymatic steps and is itself the precursor of melatonin in the pineal gland. The serotonin transporter, which clears it from synapses, is the target of SSRI antidepressants. Serotonin also occurs in plants, fungi and animal venoms, including wasp and scorpion stings.

Who wears it

Neuroscience and psychology students buy this shirt, and so do the pharmacists and psychiatric nurses who dispense the drugs that act on the serotonin system. People who manage depression or anxiety wear it as a matter-of-fact statement about brain chemistry with no slogan attached. Gastroenterologists appreciate the irony that most serotonin lives in the gut, and some wear it on that basis alone. Yoga teachers and runners like it for the association with mood and sleep. It is a natural pair with the dopamine shirt: two neurotransmitters, two people, one dinner-party explanation that both of them can deliver.

Frequently asked questions

Is the structure on the shirt chemically accurate?
Yes. It is the standard skeletal formula of 5-hydroxytryptamine: an indole ring system with a hydroxyl group at position 5 and a two-carbon chain ending in a primary amine at position 3. Serotonin has no stereocenters, so the flat line drawing is the complete structure.
What exactly is printed, and in which colors?
The skeletal structure, the name and the molecular formula are printed on the chest in a single ink color: white ink on a black tee or black ink on a white tee. There is no other text or graphic on the shirt, so the drawing reads as the design.
Is serotonin the happiness chemical?
That label is a simplification. Serotonin modulates mood, but it also regulates sleep, appetite, gut movement, blood clotting and bone metabolism, and most of it never reaches the brain. The shirt shows the molecule; it makes no promises.
How is serotonin related to psilocybin and LSD?
All three share the indole-ethylamine skeleton of tryptamine. Psilocybin and its active form psilocin are close structural cousins of serotonin and act on the same 5-HT2A receptor, as does LSD, which carries the tryptamine framework inside a larger ring system. This is why the serotonin shirt is often bought alongside the psilocybin, DMT and LSD designs.
